Excystment-Dependent Alteration of Protein Expression in Terrestrial Ciliate Colpoda cucullus
نویسندگان
چکیده
Protein expression during the excystment of Colpoda cucullus was studied by SDS-PAGE. The expression levels of 60-, 50- and 49-kDa proteins were markedly changed from the early to later stage of excystment. The 60-kDa protein (p60) was temporarily expressed first, and its expression was inhibited by actinomycin D. LC-MS/MS analysis showed that the amino acid sequences of p60 partially coincided with those of the Paramecium tetraurelia unnamed protein homologous to DEAD-box RNA helicase. These results suggest that p60 expression is enhanced by transcriptional regulation and may be involved in initiating the molecular events leading to cellular morphogenesis.

Emergence of the Terrestrial Ciliate Colpoda cucullus from a Resting Cyst: Rupture of the Cyst Wall by Active Expansion of an Excystment Vacuole
The first sign of excysting Colpoda cucullus cells is the initiation of the pulsation of a contractile vacuole, which is then replaced by a non-pulsating vacuole (excystment vacuole) that continues to expand and finally ruptures the outermost cyst wall (ectocyst) due to inner pressure. A ciliate surrounded by flexible membranes (endocyst) thus emerges. The osmolarity of the excysting cells is e...
متن کاملIdentification of cAMP-dependent phosphorylated proteins involved in the formation of environment-resistant resting cysts by the terrestrial ciliate Colpoda cucullus
In the terrestrial ciliate Colpoda cucullus, an elevation of the intracellular cAMP concentration was reported to be involved in environment-resistant resting cyst formation. In the present study, cAMP-dependently phosphorylated proteins of encystment-induced C. cucullus were isolated with Phos-tag agarose phosphate-affinity beads and subsequent SDS-PAGE. In a liquid chromatography/tandem mass ...
متن کاملThe Kinetics of Excystment in Colpoda Duodenaria
1. Extensive experimental data have been collected on the time required for the excystment process of the small ciliate Colpoda duodenaria throughout a range of temperatures of 8 degrees through 32 degrees C. and a range of concentrations of yeast extract excystment media of 0.08 through 22.4 gm./liter. 2. The excystment process has been separated into two periods, the first inversely proportio...
متن کاملA Case of Ciliate Protozoa Colpoda Spp. (Ciliata: Colpodidae) Detected In Human Urine
In the urine of a patient with chronic prostatitis, renal microlithiasis and acute cystitis we found the ciliate protozoa Colpoda spp., both in vegetative and cystic form. The entry point was most likely the urinary tract. Keeping in mind that only four more cases of Colpoda spp. existent in human urine have already been described, and that in the case of our patient the ciliate was present at ...
متن کاملNew records of the ectoparasitic flagellate Colpodella gonderi on non-Colpoda ciliates.
Colpodella gonderi is the only ectoparasitic flagellate of ciliated protozoa described thus far. This investigation reveals new records of C. gonderi retrieved from soil samples in southern Scotland, UK. Of fourteen ciliates species identified in one single occasion, three of them, Colpoda steinii, Pseudoplatyophrya nana and Grossglockneria acuta, were infested with the parasite. These results ...
